It replaced the closed Johor Bahru railway station located 200m south of it. It was inaugurated on 21 October 2010. It is connected to both the Sultan Iskandar Customs, Immigration and Quarantine Complex and City Square shopping center by pedestrian overhead bridges.

There are seven trains daily passing through Johor Bahru Sentral in each direction. The southbound trains originate at Butterworth, Gemas, Kuala Lumpur, Kuala Lipis or Tumpat in West Malaysia and terminate at Singapore's Woodlands Train Checkpoint.
